摘 要
目的观察阿托伐他汀、辛伐他汀对大鼠脑缺血再灌注血清MDA、SOD浓度和脑组织eNOS、nNOS和iNOS表达的影响,探讨他汀类药物对急性脑梗死的保护作用机制。方法36只Wistar大鼠随机分为假手术组(Sham)、缺血再灌注IR组(IR)、阿伐他汀(Ator)及辛伐他汀(Sim)预处理高、低剂量组,每组6只。高剂量组和低剂量分剐以6mg/,连续给药7天。用线栓法制备大脑中动脉缺血再灌注模型,缺血2JI,再灌注22h。用比色法检测外周血中MAD、SOD的浓度,用免疫组化法检测脑组织中eNOS、nNOS和iNOS 的表达。结果(1)辛伐他汀、阿托伐他汀预处理各组外周血SOD浓度较IR组显著升高,MDA浓度显著降低。(2)各预处理组nNOS和iNOS表达较IR组显著
降低(P<以口J,氏口彻。除辛伐他汀低剂量组以外其它各预处理组eNOS
的表达均较IR组显著增加(氏理口功。(3)在上述指标的比较中,辛伐他汀和阿托伐他汀之间差异无显著性(乃口∞,高剂量组对上述指标的影响较低剂量组有显著的趋势,但差异无显著性(伶口鳓。
结论辛伐他汀、阿托伐他汀预处理能提高大鼠脑缺血再灌注后外周血 SOD浓度、降低MDA浓度,可抑制脑组织iNOS、nNOS的表达,上调 eNoS表达。关键词脑缺血再灌注;辛伐他汀:阿托伐他汀;超氧化物歧化酶:丙二醛;~氧化氮合酶

Simvastatin and Atorvastatin Influence on Serum MDA,SOD and Expression of NOS in Rats with Focal Cerebral Ischemia-Reperfusion
ABSTRACT
Objective To observe the influences of simvastatin and atorvastatin on thickness of serum MDA,SOD and expression of NOS in rats with focal cerebral ischemia reperfosion,and probe the neuroprotective mechanisms of statins.
Methods Thirty-six Wistar rats were randomly divided into six groups: sham ischemic group(sham),ischemia-reperfusion group(IR),and simvastatin (Sim)and atorvastatin(Ator)preconditioning in high and low dose groups. High-dose and low-dose preconditioning groups were administrated Sim or Ator 6mg/kg and ,kg by douche via stomach once a day for 7 focal cerebral ischemia-reperfusion model was established in Wistar rats by using the method of thread inserting left middle cerebral artery(MCA)for 2h and reperfusing for expression of eNOS,nNOS and iNOS was
