RAC-OVA 抗原和抗 RAC 单克隆抗体的筛选

摘要:在进行酶联免疫反应实验操作过程中,为了保证材料质量及使用的延续性,在使用前分别对两对抗原抗体进行了筛选和
评价。主要是从抗体灵敏度的测定、抗体特异性的测定和抗体效价的测定这三个方面来选择灵敏度好、特异性强和效价高的一对抗原
抗体。其中抗体灵敏度的测定中,单位 1(江南大学)的抗体 IC50的平均值为 μg/L,灵敏度较好;两家单位的莱克多巴胺单克隆
抗体的特异性都较好;单位 1 的抗体效价为 1:32000,效价较高。最终选择江南大学提供的 RAC-OVA 抗原,原始浓度为 10 mg/mL,
抗莱克多巴胺单克隆抗体的原始浓度为 mg/mL。

Selection of RAC-OVA Antigen and Monoclonal anti-RAC antibody

Abstract: During the ELISA experimental operation process, two pairs of antigen and antibody with a good sensitivity,strong specificity
and high titer were screened and evaluated respectively before used in order to ensure quality of materials and use of continuity. Determination
of antibody sensitivity (antibody made by Jiangnan University) showed an average of IC50 being μg/L with high sensitivity. Ractopamine
monoclonal antibodies of two units were both good. Antibody titer of the antibody made by Jiangnan University was 1:32000,which was better
than the other. The RAC-OVA antigen made by Jiangnan university was used with original concentration 10 mg/mL. The
