目的:
探讨汉防己甲素抑制PANC-1细胞增殖作用的可能分子机制。
方法:
1. 实时定量PCR检测PANC-1细胞经汉防己甲素处理24-72 h后细胞周期相关基因
P21cip/waf1、c-myc、Cdc25A、E2F1和凋亡相关基因survivin mRNA水平变化。
2. western blot检测经汉防己甲素处理24-72 h后PANC-1细胞P21cip/waf1蛋白表达水平的
改变。
结果:
1 实时定量 PCR 结果显示,PANC-1 细胞经汉防己甲素处理 24 h,与对照组相比,
P21cip/waf1 mRNA 水平明显增加,而 Cdc25A、c-myc、E2F1、survivin mRNA 水平
均下降。
blot 结果表明 PANC-1 细胞中 P21cip/waf1 蛋白在汉防己甲素处理早期升高。
结论:
P21cip/waf1 mRNA 和蛋白的表达,下调 Cdc25A、c-myc、
E2F1 mRNA 的表达,致使 PANC-1 细胞 G1 期阻滞。
2. 汉防己甲素也可能通过下调 survivin mRNA 的表达,诱导 PANC-1 细胞凋亡的发生。

Abstract
PartⅠ The effects of tetrandrine on the proliferation of human
pancreatic cancer cell line PANC-1

Objective
To observe the effects of tetrandrine on the proliferation of human pancreatic cancer
cell line PANC-1.
Methods
was used to detect the medicine sensitivity of PANC-1 cells to different
concentrations of tetrandrine.
-induced cell cycle arrest was detected by flow cytometry.
cytometry was used to detect the effect of tetrandrine induced apoptosis of PANC-1
cells .
Results:
results showed that tetrandrine elicited a concentration- and time- dependent manner
inhibiting proliferation of PANC-1 cells with an IC50~17 and~10 μmol/L for 24 h and
72 h exposures in MTT assay. The maximum growth inhibition rate of tetrandrine was
100%, which occurred at 72 h using 80 μmol/L tetrandrine.
2. G1 phase cell cycle arrest of PANC-1 cells were significantly induced after tetrandrine
treatment 24 h (% cell population in G1 when treated by 20 μmol/L tetrandrine). In
contrast, cell population in S phase was significantly decreased to % at the same
treatment. Cell population in G2/M phase can increase to % in the condition of 20
μmol/L tetrandrine treatment.
