【摘要】目的探讨BP1基因在急性白血病患者中的表达及其临床意义。方法运用逆转录-聚合酶链反应(RT-PCR)方法检测HEL白血病细胞系(BP1阳性细胞株)、92例急性白血病(AL)患者及10名正常对照者BP1基因的表达水平;分析其在急性白血病中的表达规律及与临床预后的关系。结果 10名正常人和急性淋巴细胞白血病患者的骨髓细胞中无BP1基因表达(阴性);HEL细胞系BP1阳性;急性髓系白血病(AML)%,在各亚型AML中均表达,AML各亚型间的阳性率无差异()。AML初治、复发、%、%、%,各组间无明显差异()。AML初治患者中BP1阳性表达者首次完全缓解(CR1)后缓解持续时间短于BP1阴性表达者(),早期病死率(确诊后3个月内死亡)明显升高()。结论 BP1基因在AML中有异常高表达,具有髓系特异性,可作为区别淋系和髓系的标志性基因。BP1基因在AML的发病中可能起一定作用,可作为判断AML预后的一个指标。

Abstract: Objective To investigate the expression of BP1 gene in patients with acute leukemia and its significance. Methods BP1 level was determined by reverse transcription polymerase chain reaction (RT-PCR) in HEL leukemia cell line in 92 patients with acute leukemia and 10 healthy volunteers. The regularity of the expression of BP1 gene in acute leukemia and the relationship between BP1 level and prognosis of leukemia was explored. Results BP1 mRNA was not expressed in 10 healthy volunteers and patients with acute lymphoid leukemia, while it was positive in HEL cell line. The total positive rate of BP1 gene expression in AML was %. BP1 gene was expressed in all subtypes of acute myelogenous leukemia, with no marked difference (). The positive rate was % for the newly diagnosed, % for those with relapse and % for those plete remission. There were no significant differences in positivity among groups (). The remission phase after CR1 was shorter in BP1 (+) patients than in the (-) ones, and the early mortality rate was higher in the former than in the latter (). Conclusion BP1 was abnormally overexpressed in acute myelogenous leukemia, which suggests that it is myelogenous specific and could serve as a marker gene to distinguish myelogenous and lymphoid genes.
