【摘要】目的 E2蛋白在自发性高血压大鼠(SHR)心脏不同部位的分布和表达差异,及鼠龄对其表达水平的影响。方法提取幼龄和高龄SHR心肌不同部位(心室、室隔膜和心房)组织的膜蛋白,利用Western印迹技术和图象分析方法,E2蛋白在样品之间免疫强度差异。结果 E2蛋白的分布;E2蛋白的表达均无显著性差异;但在高龄大鼠的右心室和室隔膜组织中,KCNE2蛋白的表达显著高于其在幼龄大鼠相同组织中的表达。结论 KCNE2蛋白在SHR心脏不同部位均有分布;在高龄大鼠右心室和室隔膜组织中,KCNE2蛋白的表达水平均高于其在幼龄大鼠同一组织中的表达。

ABSTRACT: Objective To investigate the distribution and expression of KCNE2 in different parts of myocardium of spontaneously hypertensive rats (SHR) pare differences of KCNE expression in cardiac muscle between young and old SHR. Methods First membrane proteins were prepared from right ventricular myocardium(RV), interventricular septum, left ventricular myocardium(LV) and atrial myocardium from young and old SHR, respectively. Then the membrane proteins were separated by SDS
PAGE, transferred to polyscreen PVDF membrane, probed with a primary antiKCNE2 (polyclonal Ab), and visualized with ECL detection kit. Images were acquired and analyzed with related software. Results KCNE2 protein was found to distribute in all different parts of myocardium of SHR with no significant differences. The expression of KCNE2, however, in right ventricular and interventricular septum of old rats was significantly higher than that of young rats. Conclusion KCNE2 protein distributes in all different parts of myocardium of SHR. The expression of KCNE2 in right ventricular and interventricular septum of old rats is significantly higher than that of young rats.

KCNE2蛋白也称MiRP1(minkrelated protein 1)蛋白,是一个含有123个氨基酸、具有一个跨膜结构域的蛋白[1]。KCNE2蛋白是几个电压门离子通道的
β亚基,可与多个离子通道的a亚基如HERG通道、、KCNQ1通道和超极化激活的阳离子通道相互作用,调节和影响着这些离子通道的功能[13]。KCNE2蛋白很可能是心脏电生理的一个重要的调控子[2],该亚基可以和HERG通道的α亚基或/Q1亚基联合聚集形成传导IKr(快激活钾延迟整流电流)和/或IKs(慢激活钾延迟整流电流)的通道[45]。
自发性高血压大鼠(SHR)是高血压和心血管病研究的常用动物模型[67]。资料表明,心脏组织的细胞膜钾通道各亚基的分布和表达水平对心脏电生理的功能有一定的影响和调控作用[8]
