【摘要】目的检测脑缺血再灌注后不同时间点轴突生长抑制因子NogoA在损伤白质区的基因表达,探讨NogoA与缺血再灌注白质损伤的可能关系。方法制备大鼠大脑中动脉缺血再灌注(MCAO)模型,实时定量聚合酶链反应方法(Relative Quantification PCR,RQPCR)检测各时间点NogoA在损伤白质区域的基因表达。结果在损伤白质区域,NogoA基因表达在缺血再灌注1 d时表达量开始减少(与假手术组相比,),7 d时降至最低,14~21 d表达量开始上调,21 d时仍低于假手术组水平(与假手术组相比,)。结论缺血再灌注损伤后白质区域的NogoA基因表达呈现动态变化规律,针对NogoA进行干预有望为缺血再灌注损伤后的脑保护治疗找到新的靶点。

【Abstract】 Objective To study the expression of NogoA in cerebral white matter after focal cerebral ischemia
reperfusion(I/R) in rats, and explore the relationship between NogoA and cerebral white matter lesions after ischemia. Methods SpragueDawley male rats were subjected to 2 h of middle cerebral artery occlusion (MCAO). Relative quantification PCR(RQPCR) was performed to examine the expressions of NogoA gene at different reperfusion time. Results The expression level of NogoA gene in the white matter was decreased in 1 d group( vs sham group),and was the weakest in 7 d group. It was increased in 14~21 d group, and in 21 d the expression level was still lower than that of sham group(). Conclusions NogoA expreesion has dynamic changes in the white matter after focal cerebral I/R. NogoA could be as one of the targets for neuroprotective therapy after cerebral ischemia.
