【摘要】目的探讨白藜芦醇在体外对宫颈癌Hela细胞VEGF表达的影响。方法 MTT法检测不同浓度( 0、、25、50、100、200、300、400、600 μmol/L) Res处理24、48、72h对HeLa细胞的抑制率;RT-PCR方法检测VEGFmRNA表达水平,Western Blot方法检测细胞内VEGF蛋白表达水平。结果白藜芦醇呈时间剂量依赖性抑制宫颈癌Hela细胞的增殖;宫颈癌Hela细胞高水平表达VEGF,白藜芦醇能显著降低宫颈癌Hela细胞VEGF mRNA和蛋白表达。结论白藜芦醇可以下调宫颈癌Hela细胞VEGF的表达,抑制宫颈癌细胞的增殖。

【Abstract】Objective To investigate whether resveratrol could inhibit the vascular endothelial growth factor (VEGF) expression of human cervical cancer Hela cells in vitro. Methods Proliferation inhibitory effect was tested by MTT assay; VEGF mRNA was measured semi-quantitatively by RT-PCR, and the protein level was deteced by Western Resveratrol could inhibit the proliferation of Hela cells, with dose and time-depend effects; The level of VEGF was siginificantly decreased on Hela cells after incubated with resveratrol. Conclusion Resveratrol could inhibit proliferation and
lower the expression of VEGF in human cervical cancer Hela cell line in vitro.

白藜芦醇(Resveratrol, Res)是一种广泛存在于葡萄、花生等植物中的植物补体,属于非黄酮类多酚化合物,具有调节脂蛋白代谢、脂氧化、抗炎及抗肿瘤等作用[1], 且无不良作用, 是目前认为最有希望的天然抗癌药之一。研究显示, 白藜芦醇对前列腺癌、乳腺癌、肝癌、肺癌、肠癌、甲状腺癌、垂体腺瘤和血液肿瘤等多种肿瘤具有一定的防癌和抗癌活性[2,3]。本实验旨在探讨 Res对人宫颈癌Hela细胞的增殖抑制及VEGF的表达下调作用,为宫颈癌的临床治疗提供新的理
