摘要:目的利用本室构建的反义VEGF165 (血管内皮生长因子,vascular endothelial growth factor)真核表达载体,研究VEGF反义RNA表达对人胃癌细胞生物学表型的影响. 方法用阳性脂质体介导的基因转染技术,将VEGF反义RNA重组体转入人胃癌细胞系,观察转染细胞的细胞周期、增殖能力及致瘤生长情况等生物学表现. 结果 VEGF反义RNA重组体转染胃癌细胞后,斑点杂交、间接免疫荧光等分析显示,反义RNA基因转染技术可使VEGF的表达明显减弱;与未转染的细胞相比,G2 /M期细胞增加(),而S期细胞减少();克隆形成率(%)低于未转染细胞(%);肿瘤细胞接种裸鼠后33d时,VEGF反义RNA表达人胃癌细胞所致的移植瘤体积(345±136)mm3 明显小于未转染细胞所致的移植瘤体积(1534±363)mm结论VEGF反义RNA可以通过抑制肿瘤组织血管生成而防治肿瘤的生长,另外VEGF的表达可能与细胞增殖能力有关.

Abstract:AIM To investigate the effect of vascular en-dothelial growth factor(VEGF)antisense gene on the biolog-ical characteristics of human gastric cancer cells by antisense nucleotide The VEGF165 antisense gene binant was introduced into gastric cancer cell line by lipofect transfection the cell cycle,prolif-erative abiliyt in vitro and growth in nude mice of transfected cells were Introduction of the VEGF165 antisense construct into SGC-7901cells resulted in a marked reduction in the expression of VEGF-specific mRNA and protein by dot blot and immunofluorescence staining analysis in transfected tumor cycle analysis showed pared with the parental cells,the number of trans-fected cells had an increase()in G2 /M phase,a decrease()in S phase,and its colony forming rate(%)was lower than that(%)of the control volume of tumor(345±136)mm3 in the nude mice inoculated with the cells transfected by antisense VEGF165 gene construct was greatly reduced parison with that(1534
±363)mm3 in the nude mice inoculated with SGC-┐SION As repressing angiogesis,antisense VEGF gene transfection might inhibit the growth of solid tumor;in addi-tion,it might correlate with the proliferation of tumor cells.
