Effect of urokinase on cell proliferation and apoptosis of rat pleural fibroblasts
【Abstract】 AIM: To study the effect of urokinase on the cell proliferation and apoptosis of rat pleural fibroblasts. METHODS: Rat pleural fibroblasts were primarily cultured to 4-12 passages, and then the medium was added with urokinase of different concentrations. After another 24-96 h culture, light microscope was used to observe morphological change. Apoptosis bodies were examined by fluorescent staining. Flow cytometry was used to detect apoptotic cells. RESULTS: Urokinase decreased the number of viable cells in time and concentration dependent manners. No apoptosis body or apoptotic cells were found by flow cytometry. CONCLUSION: Antiproliferative effect of urokinase on rat pleural fibroblasts is confirmed, but not related to cell apoptosis.

【摘要】目的:研究尿激酶(UK)对大鼠胸膜成纤维细胞(FB)增殖与细胞凋亡的影响. 方法:将大鼠胸膜FB原代培养,4~12代用于实验,FB与不同浓度UK共培养24~96 h;用光学显微镜观察其形态学改变;荧光染色查找凋亡小体;流式细胞技术观察有无凋亡细胞出现. 结果:UK以时间和浓度依赖的方式抑制细胞生长,但未发现凋亡小体,流式细胞技术没有检测到凋亡细胞. 结论:UK能够抑制FB增殖,其机制与细胞凋亡无关.

胸膜纤维化是难治性胸膜炎的病理基础,胸膜成纤维细胞(fibroblasts, FB)增殖又是胸膜纤维化的主要原因. 目前,临床上应用尿激酶(urokinase, UK)防治炎性胸膜炎形成胸液包裹、胸膜黏连、肥厚等获得了明显效果,但机制不甚明确[1-3]. 因此,我们以研究大鼠胸膜FB为治疗靶位,观察UK对大鼠胸膜成纤维细胞增殖的抑制作用,探讨作用机制, 为UK防治炎性胸膜炎提供理论依据.
