摘要:目的:探讨 Hedgehog(Hh)信号通路抑制剂对人胃癌 BGC823 细胞增殖与凋亡作用研
究。方法:Cyclopamine(环巴明)特异性阻断 Hh 信号通路后,联合应用 Gli-1 特异性阻滞
剂 GANT61,采用 MTT 法检测细胞存活率,Annexin V-FITC/PI 双染流式细胞术检测其对细
胞凋亡的影响,Hochest33258 观察凋亡细胞核形态学改变。结果:MTT 检测 GANT61 和
Cyclopamine 的半数抑制浓度分别为 , 。联合应用 GANT61 后,流式细
胞术检测 BGC823 细胞的凋亡率明显增加。Hochest 实验结果显示,联合给药组凋亡细胞核
比例较单独给药组高(p<)。结论:阻断 Hh 信号通路能显著抑制 BGC823 细胞的增殖,
促进细胞凋亡。GANT61 联合应用 Cyclopamine 可加强胃癌细胞的凋亡,为靶向治疗胃癌提
供一种新的治疗思路与决策。
