摘要:目的:研究 CART 肽对脑缺血再灌注急性期脑水肿的作用及时间窗。方法:采用线
栓法复制雄性小鼠大脑中动脉阻塞(MCAO)和再灌注模型。实验分 Sham 组、MCAO 组、
四个 CART 治疗组( 0h、2h、3h 和 4h 组)。通过 TTC 染色检测脑梗死体积及肿胀体积,
应用干湿重法测定脑水含量,伊文思蓝法检测血脑屏障(BBB)的损坏程度。结果:MCAO
组小鼠脑梗死体积、肿胀体积、脑水含量和伊文思蓝漏出率都明显高于 Sham 组,CART 0h
组的脑梗死体积、肿胀体积、脑水含量和伊文思蓝漏出率明显低于 MCAO 模型组。CART 2h、
3h 组脑肿胀体积明显低于 MCAO 组,CART 4h 组脑肿胀体积与 MCAO 组没有明显差异。
结论:(1)CART 肽能减轻脑缺血再灌注引起的脑肿胀和脑水含量,抑制血脑屏障的破坏;
(2)再灌注后 0h 给予 CART 肽作用明显,2h 和 3h 给予 CART 肽仍然有效。
