摘要:尽管肿瘤坏死因子受体相关因子 4 在很多肿瘤中起到了重要的作用,但其具体的作用
机制还并不清楚。在本实验中,我们运用免疫印记的方法分析了 TRAF4 的表达,结果显示
TRAF4 总蛋白表达在人正常乳腺细胞系 MCF-10A 中的表达低于乳腺癌细胞系 MCF-7 及
MDA-MB-231。TRAF4 核蛋白表达在 MCF-10A 中显著高于 MCF-7 及 MDA-MB-231;浆蛋
白表达显著低于 MCF-7 及 MDA-MB-231。TNF-α处理后,MCF-7 细胞中转染 siRNA-TRAF4
使其表达沉默可抑制该细胞增殖,且 NF-κB 的核表达显著降低;MCF-7 细胞早期凋亡细胞
数显著增加。我们的结果说明了在 MCF-7 细胞中,TRAF4 在 TNF-α诱导的细胞凋亡通路
中起抗凋亡作用。

The expression and anti-apoptosis function of TRAF4 in

Abstract: Although numerous evidences show that Tumor Necrosis Factor Receptor-Associated
Factor 4 (TRAF4) plays an important role in many tumors, the exact molecular mechanism
remains elusive. In the present study, we analyzed the expression of TRAF4 with
immunofluorescence and western blot. We found that the expression of TRAF4 in normal breast
cells MCF-10A was lower than that in breast cancer cells MCF-7 and MDA-MB-231. TRAF4
localized in both cytoplasm and nuclei in MCF-7 and MDA-MB-231. After TNF-α treatment,
TRAF4 depletion by siRNA in MCF-7 cells could suppress cell proliferation and the nuclear
