文档介绍:摘要本文设计正交试验,确定了水提醇沉法提取了兰州百合粗多糖的最佳工艺条件为:温度90℃,时间2h,料液比1:10(w/v)。同时对脱蛋白方法进行了研究,结果表明酶和Sevage法联用适合兰州百合多糖的脱蛋白。用葡聚糖凝胶 ,收集主峰多糖后得到一种多糖一BHI,相对含量为 %。本文采用光散射法(动静态光散射仪)、比旋光度法(自动旋光仪)热分析仪等研究了BHI的物理性质。用光散射法测定其重均分子量为5。17×1049/tool, 比旋光度(a]”。。(,H:o),%,显色反应表明BHI 是还原性多糖,热分析测试为无定型粉末。通过纸色谱、、、D-半乳糖等三种六碳糖组成,其组成摩尔比为10:19:1。本文首次根据BHI糖***衍生物的 GC分析、BHI高碘酸氧化分析、BHI***化、ⅡL13C-NMR,1H-NMR, ,确定了BHI中单糖的糖苷键类型、分支点及连接方式如下: BHI多糖以8-Glu(1—4)(1—3)以接近2:l的比例交替形成主链,(1—6),(1,3) 位有分枝,还原性末端为B—Glu(1一)。采用原子力显微镜,首次观察了BHI的空间形貌,结果表明BHl分子为岛屿状线团,浓度增大时,分子之间相互交联,呈现网络状结构。关键词:兰州百合, 多糖, 化学结构, 空间形貌 Abstract Through orthogonal experiment we know that theoptimum extractionLilium davidii Salisb crude haride condition ofhot-water and alcohol-precipitation method isthetimeof2h,the tempemturc of90"(2and the material/extraction solution ratio of1:10(w/v).Among the three methods of elimination protein,the best istheenzyme-scvage new kind of haride,named BHl which relative content was %,was purified from haride bysephadex G-100 column.
