姜黄素对人卵巢癌细胞株 SKOV-3 增殖和凋亡的影响
摘要
目的
探讨姜黄素对体外培养人卵巢癌 SKOV-3 细胞增殖和凋亡的影响。
方法
选用人卵巢癌细胞株 SKOV-3 进行体外培养,以 5~80μmol/ L 姜黄素分别处理 SKOV-3
细胞 24~72h,MTT 法检测细胞的生长抑制率;流式细胞仪检测细胞周期;SABC 法检测
survivin、caspase-3 在细胞中的表达。
结果
姜黄素能显著抑制 SKOV-3 细胞体外生长,呈时间剂量依赖性。姜黄素作用 24h 随浓
度的升高 survivin 阳性表达逐渐降低,caspase-3 阳性表达逐渐升高。流式细胞仪分析证
实姜黄素能使 SKOV-3 细胞积聚在 S 期和 G2/M 期。
结论
姜黄素对人卵巢癌细胞株 SKOV-3 的生长有显著的抑制作用,survivin 的表达下降、
caspase-3 的表达上调可能参与了诱导细胞凋亡。

Effects of curcumin on proliferation and apoptosis of human
ovarian cancer cell line SKOV-3 in vitro
Abstract
Objective
To investigate the proliferation and apoptosis effect of curcumin on human ovary cancer cell
line SKOV-3 in vitro.
Methods
Human ovarian cancer cell line SKOV-3,cultured in vitro,was treated with 5~80μ mol/L
curcumin for 24~ MTT method was used to measure growth inhibition rates of SKOV-3
cells. The cell cycle distributions were detected by flow cytometic expression of
survivin and caspase-3 in SKOV-3 cell was observed by SABC immunohistochemistry.
Results
Curcumin could effectively inhibit the in vitro growth of human ovary cancer cell line
SKOV-3 in time- and dose-dependent manners. When treated with curcumin for 24 hours, the
expression of survivin was decreased while the expression of caspase-3 was increased. The cell
growth was arrested at S and G2/M stage was observed by FCM.
Conclusion
Curcumin could significantly inhibit the growth of ovary cancer cell line SKOV-3 in vitro. The
apoptosis is related to the down-regulation of survivin and up-regulation of caspase-3.
