负载供肾抗原的DC2细胞延长大鼠移植肾存活观察

摘要:【目的】 检测NAD(P)H氧化酶及ROS在不同恶性程度胶质瘤中的表达,探讨NAD(P)H氧化酶及细胞内ROS对胶质瘤存活、增殖、凋亡以及恶性表型的影响.
观察负载供肾大鼠抗原的DC2(未成熟DC)在体内特异性抑制急性排斥反应及其对移植物的功能和存活时间的影响。【方法】体外制备并诱导分化未成熟的优势淋巴系DC前体细胞(DC2),进行免疫表型分析,健康成年BN大鼠和Lewis大鼠各57 只,BN大鼠为供者,Lewis大鼠为受者,随机分为4 组,术前负载供肾大鼠抗原和无关供肾抗原,观察DC2体外抑制混合淋巴细胞反应(MLR)的情况,构建大鼠同种异体肾移植模型,观察其在体内特异性抑制移植排斥反应发生的情况,Kaplan-Meier生存分析研究对移植物的存活时间的影响。【结果】所制备的DC2经供肾大鼠抗原负载对MLR具有抑制作用(P〈0。05),负载供肾抗原DC2输注的大鼠肾移植术后中位生存时间是62±5。53 天,负载无关大鼠抗原和不负载抗原输注后移植实验组,术后中位生存时间分别是30±3。95 天和27±4。74 天,实验组与对照组血清肌酐,炎症因子IL-4和IL—10等表达,差异具有统计学意义(P〈).【结论】大鼠同系受体来源的负载供肾抗原的DC2具有体外抑制MLR的作用,能够预防和抑制急性排斥反应的发生,延长生存时间,提高远期存活率。个人收集整理,勿做商业用途本文为互联网收集,请勿用作商业用途

Abstract: 【Objective】To observe the specific inhibition to the acute rejection of the rat kidney antigen conditioned DC2 (immature DC) in vivo and its effect on graft function and survival time. 【Methods】To prepare in vitro immature precursor lymphoid DC (DC2), analysis the immunophenotype, 57 adult BN rats and Lewis rats were separately recruited into experiment,BN rats are donor, Lewis rats are recipient,they were randomly divided into 4 groups。 Then condition the DC2 with donor rats kidney and unrelated kidney antigens, observe its inhibition of mixed lymphocyte reaction (MLR ) in vitro, construct the allogeneic rat kidney transplantation model of, observed specific inhibition of allograft rejection in vivo, use Kaplan—Meier method to analysis survival time of graft。 【Results】The rat kidney antigen conditioned DC2 inhibited MLR (P<0。05), the median survival time of the rats conditioned with the donor rats kidney antigen after renal transplantation were 62±5。53 days, but that of the independent kidney antigen conditioned or non-antigen conditioned groups were 30±3。95 and 27± days。 The peripheral blood creatinine, inflammatory cytokines IL—4 and IL—10 expressions of the experimental groups was statist
