摘 要:以小麦稳定表达的微管蛋白基因(Tubulin)作为对照,利用半定量反转录聚合酶链式反应(Sem i-QRT-PCR)技术对与抗叶锈病有关的小麦(T riticum aestivum L. ) ATG8基因的表达进彳亍了研究。结果 发现在叶锈菌诱导不同时间后,ATG8基因在小麦品种5389叶片中的表达量基本不变;在小麦品种洛 夫林10叶片中表达量出现变化,在接种后16h ATG8基因表达量开始上升,在48h有所下降, 之后又被诱导表达。研究表明,用半定量RT-PCR技术研究小麦基因表达,具有特异性高、操作简便 和可靠性强的优点。

Express ion Analys is of Wheat ATG8 Gene by Sem i-QRT-PCR
Abstract: The stable expression of wheat microtubule protein (Tubulin) as control, using semi-quantitative reverse transcription polymerase chain reaction technique (Semi-QRT-PCR), and the wheat leaf rust resistance (T riticum aestivum L. ) ATG8 gene expression were studied. The results showed that the leaf rust induced at different times, the ATG8 gene expression in the wheat varieties 5389 leaves basically unchanged; in the wheat varieties Lovrin 10 leaves changed, after vaccination at 16h ATG8 gene expression level began to rise, at 48h declined ,after be induced to express results show that semi-quantitative RT-PCR gene expression in wheat technology, with high specificity, simple operation and reliability advantages.
