ts: The PC-3 was initiated from a bone metastasis of a grade IV prostatic adenocarcinoma
from a 62-year-old male Caucasian. The cells exhibit low acid phosphatase and
testosterone-5-alpha reductase activities.
Propagation: Complete growth medium: RPMI1640+10%FBS+P/S
Temperature:
Atmosphere: air, 95%; carbon dioxide (CO2), 5%
Subculturing: Protocol:
1. Remove and discard culture medium.
2. Briefly rinse the cell layer with % (w/v) Trypsin- mM EDTA solution
to remove all traces of serum that contains trypsin inhibitor.
3. Add to ml of Trypsin-EDTA solution to flask and observe cells under
an inverted microscope until cell layer is dispersed.
Note: To avoid clumping do not agitate the cells by hitting or shaking theflask while waiting for the cells to detach. Cells that are difficult to
