Codon Usage Bias in the Chloroplast Genome of Asparagus Officinalis

Abstract Based on the composition and characteristics of chloroplast (cp) genomic codon in Asparagus
officinalis L., we analyzed its codon usage bias (CUB) preferred use mode in order to provide a basis for further
genetic research. In this study, the effective number of codons (ENC), relative synonymous codon usage (RSCU),
ENC plot value and optimal codon of 52 coding gene sequences in cp genome were analyzed by Bioinformatics
and statistical methods. The results showed that the average GC content of codon bases 1, 2 and 3 in cp genome
showed a decreasing trend (GC1 (%)>GC2 (%)>GC3 (%); the size of ENC value ranged from
to , of which 42 ENC values were larger than 45; the RSCU value of 28 codons was larger than 1,
mainly ending with AT, and this bias was weak; Neutral mapping analysis showed that the correlation between
GC12 and GC3 was low (r=, P>); ENC plot analysis showed that there were differences in the
frequency of base use (U>A, G>C); And 16 base combinations (CGU, GGU, UUA, UUG
