Supported by the National Natural Science Foundation of Chphase during carcinogenesis were obtained: 25 hyperplasia, 27 squamous metaplasia, 37 dysplasia, 30 carcinoma in situ and 25 infiltrating carcinoma. The positive rates of p16 and p27 proteins decreased and the positive rate of p53 protein increased obviously when the carcinoma developed. The expression of p16 was positively correlated with p27 (P < ). The expression of p53 was negatively correlated with p16 and p27 (P < ). The loss of p16 and p27 expression, p53 overexpressions were frequently found in lung carcinoma and there was very significant statistical difference between carcinoma and corresponding adjacent noncancerous tissue or normal tissue (P < ). Conclusion The decreased expression of p16, p27 and increased expression of p53 in the process of carcinogenesis may be associated with the genesis and development of lung squamous cell carcinoma in rats.

肺癌是全球性的恶性肿瘤[1],其发病机制一直不清。研究肺癌发生的分子机制,尤其是癌变过程的分子机制,对于提高肺癌早期诊断,降低肺癌发病率和死亡率无疑具有重要意义[2]。细胞生命活动以细胞周期为中心,肿瘤发生、发展的研究,都涉及到细胞周期及其调控。细胞周期调控的异常是肿瘤发生的一个重要前提,主要表现在细胞周期分布时相的变化和相关调控因子-细胞周期蛋白(cyclins)、细胞周期蛋白依赖性激酶(CDKs)及其抑制剂(CDKIs)表达异常, 因此不能正常调控细胞的周期性变化,从而导致肿瘤细胞的恶性增殖。p16、p27和p53是目前发现的与肿瘤细胞周期调控最密切的几种抑癌基因,但是目前关于它们在肺癌癌变过程中动态表达及其相关关系的报道较少。本研究在我室前期建立的动物模型的基
