题 目 : 声动力治疗巨噬细胞通过促进ATP释放趋化单核细胞 f subjects: The control group(macrophages alone),the SDT group, the zVAD—fmk group(added caspase-inhibitor zAVD-fmk ) and the SDT—zAVD group(like the SDT group pretreated with zAVD—fmk).Supernatants and cell debris after high speed centrifugation from these four groups were investigated respectively.
Results: Cell—free supernatants with SDT were assessed for their ability to attract THP-1 monocytes in a transwell migration assay. The SDT group induced an approximately sixfold greater recruitment of monocytes than the control group did(p<), while the monocytes migration of the SDT—ATPase group decreased significantly compared to the SDT , there was no statistic difference between the control group and the ATPase group(p>)。 Luminometer was used to measure the change of ATP ATP concentration of the SDT group was increased about therefold in comparison with the control group (p〈),and the SDT-zAVD group had a distinct decrease compared with the SDT group。 The control group and the zVAD—fmk group showed no statistic difference(p〉)。
Conclusions: This study demonstrates that SDT could induce the recruitment of monocytes by promoting the release of ATP in THP-1 macrophages via caspase—dependent pathway .

动脉粥样硬化(Atherosclerosis,AS)是一种多因性疾病,近年来,尽管AS的诊断与治疗有了长足的进步,,如血栓形成学说、脂质浸润学说、单克隆学说、损伤反应学说、剪切应力学说、同型半胱氨酸学说、精氨酸学说、内皮功能紊乱学说、氧化应激学说等都不足以完全解释AS的发病基础[1]。近年来的基础研究与临床研究结果提示,,在AS性疾病的不同临床表现形式中,炎症参与其发生和发展的所有环节[2].这一发现,为临床治疗AS提供了理论依据及研发新药提供了新思路.
1。 巨噬细胞凋亡与动脉粥样硬化
近年的研究表明,动脉粥样硬化病灶内脂质中心的大小、炎细胞尤其是巨噬细胞的数量、纤维帽的厚薄及新生血管的多少是决定斑块稳定与否的最主要的因素,而巨噬细胞则通过对斑块内脂质含量、炎症反应、纤维成分的降解及新血管形成等方面来影响动脉粥样硬化病变的进展,从脂质条纹的发生到最终斑块破裂和血栓形成,巨噬细胞在动脉粥样硬化的所有阶段都起着极其重要的作用.
