2．临床基因扩增诊断实验室质量保证
临床基因扩增诊断实验室质量保证涉及到整个基因扩增检测的所有阶段，即测定分析前的标本来集处理、测定中的核酸提取、扩增和产物分析以及测定后的结果报告等。
2．1．标本的采集
常用于基因扩增检测的临床标本包括EDTA或拘檬酸钠抗凝全血或骨髓、血清或血浆．痰、脑普液、尿及分泌物等。采样容器最好是密闭的一次性的，如真空系血管。一次性塑料容器使用时无需进一步预处理。当使用非密闹采样系统时，如尿、分泌物和骨髓的采禅，必须注意防止来自来样者的皮屑或分泌物的污染。采样者采样时起码要谈．一次性手套。可重复使用的玻璃器〔应高压处理，因为玻璃器（常含有不易失活的ANA酶。RNA酶的主要来源是实验人员的手。最好是热灭菌， 250 烘烤 4小时以上可使RNA酶永久性失活。全血和骨髓标本必须进行抗凝处理。通常EDTA和拘檬酸盐是首选的抗凝剂。不能使用肝素抗凝，因为肝素是Taq酶的强抑制剂，而且在其后的核酸提取步骤中很难去除肝素。临床用于RNA（如HCV RNA）测定的血标本最好进行抗凝处理，并尽快（3小时以内）分离血浆，以避免RNA的降解。如未作抗凝处理，则物血后，必须在1小时内分离血清。
