摘要
牡丹种子产量高,含油量高达27%,且籽油中含有大量的不饱和脂肪酸(>90%),其中油酸约占21%,亚油酸(ω-6脂肪酸)约占26%,α-亚麻酸(ω-3脂肪酸)高达45%。牡丹籽油可以作为一种可持续利用的食用油新资源。尽管如此,大多数的牡丹研究者都致力于牡丹的花芽发育、休眠、二次开花以及切花保鲜等研究,而对不饱和脂肪酸合成机制的研究才刚刚起步。油用牡丹的优良品种“凤丹白”生长繁殖快,且种子含油率和亚麻酸含量均较高。
因此,本研究以牡丹“凤丹白”为材料,提取了牡丹不同发育期种子的RNA,经DNase消化后逆转录合成cDNA。根据文献报道的牡丹种子转录组序列设计引物,以种子cDNA为模板,克隆了SAD基因并测序鉴定。本论文的研究结果将为研究油用牡丹不饱和酸合成的分子机理奠定了基础。

Cloning of SAD gene from peony "Feng Dan Bai" in oil
ABSTRACT
Peony high seed yield, oil content of up to 27% and oil containing a large amount of unsaturated fatty acids (90%), which accounts for about 21% of oleic acid, linoleic acid (omega-6 fatty acid) accounted for about 26%, alpha linolenic acid (omega-3 fatty acids) as high as 45%. Peony seed oil can be used as a new resource for sustainable utilization of edible oil. In spite of this, most of the peony researchers are devoted to the study of the flower bud development, dormancy, the two flowering and the preservation of cut flowers, and the study on the mechanism of the synthesis of unsaturated fatty acids is just started. Oil with fine varieties of peony "Feng Dan Bai" fast growth and reproduction and seed oil content and linolenic acid content were higher.
Therefore, the research to peony "Feng Dan Bai" as the material extracted RNA of peony seeds at different developmental stages, the DNase digested cDNA was synthesized by reverse transcription. According to the report of the tree peony seed transcription group, the primers were designed, the SAD gene was cloned and sequenced and identified with the seed cDNA as the template. The research results of this paper will lay the foundation for the study of the molecular mechanism of the synthesis of unsaturated acid in oil.
