达比加群酯的临床药理及其预防心房颤动患者卒中的研究进展

[摘要] 达比加群酯是近几年上市的一种新型口服抗凝药物,最初用于预防关节置换术后血栓的形
成。美国 FDA 于 2010 年 10 月批准达比加群酯用于预防心房颤动患者的脑卒中。本文对其药理作用、药代
动力学、临床疗效评价及不良反应等做一综述。

Dabigatran etexilate,a novel oral anticoagulant for stroke prevention
in patients with atrial fibrillation

[Abstract] Dabigatran etexilate,a novel oral direct thrombin inhibitor that was firstly used in preventing ve-
nous thrombo-embolism after arthroplasty,has been proved by the American Food and Drug Administration ( FDA)
for stroke prevention in patients with atrial fibrillation in October 2010. In this paper,we reviewed the pharmacolo-
gy,ics,clinical efficacy and safety of dabigatran etexilate.

达比加群酯( dabigatran etexilate,商品名: Prada-
xa) 是 2008 年首先在德国和英国上市的一种新型口
服抗凝药物,由德国勃林格殷格翰( Boehringer In-
gelheim) 公司研发,属于非肽类凝血酶抑制剂。其
化学名称为 3-[2-[4-[( 己氧基) 羰基]氨基]亚氨甲
基]苯基]氨基]甲基]-1-甲基-1H-苯并咪唑-5-基]
羰基]( 吡啶-2-基) 氨基]丙酸乙酯,结构式见图 1。图 1 达比加群酯的结构式
