Cassia seed is monly used traditional Chinese herb. It was firstly recorded in Shen Nong Ben Cao Jing and was classified as a top grade drug. At present, it has been recorded in Chinese Pharmacopoeias as the dry and ripe seed of Cassia obtusifolia Cassia tora L..
In recent years the demands of cassia seed is increasing in both international and domestic markets. This work is one branch of the basic platform projects called“ Standardization, integration and sharing pilot projects of Medicinal plant germplasm resources”, which is supported by National Science and Technology Ministry of Science and Technology . The quality of Cassia obtusifolia L. had been investigated and evaluated in our early work. The further research on the quality of Cassia tora the parison between Cassia obtusifolia L. and Cassia tora L. had been carried out in this paper. The testing protocol and quality standard of Semen Cassiae have been formulated as well. We have achieved the following fruits and innovation:
We collected 50 samples from 21 counties in 3 provinces for the first time, and formulated the Description and Data Standard for Cassia tora L.. We described the biological characteristics of each germplasm resources including 96 items. We discovered that each germplasm resource differs in many aspects such as plant morphology, medicinal characters and position. The deviation of germplasm may cause the difference in the quality of drugs.
We also investigated the control article and the developing solvent system of TLC for the identification test of cassia seed. In the basis of Cassiae Torae TLC in China Pharmacopoeia 2005 version, added aurantio-obtusin and emodin as control article for the first time, we performed TLC of cassia seed with the control articles of emodin, chryso, phano1 physcione and aurantio-obtusin. The best developing solvent
system is the mixture of petroleum ether, acetidin and formic acid(15:5:1).The
chromatographic spot is more pronounced
