【摘要】 目的研究红豆杉枝叶中紫杉醇的含量测定方法,并对人工种植的云南红豆杉中紫杉醇的含量进行调查研究。方法采用甲醇和二***甲烷超声提取样品,HPLC梯度洗脱。结果紫杉醇的理论塔板数为5628,,。回归方程为Y=4×107X+39 942,r= 7, 2~ 1/ml之间的线性关系良好。%,%(n=5),,%,%,%,RSD(n=5)%,%,%。人工种植的云南红豆杉中紫杉醇的平均含量达140mg/kg。结论该方法缩减了红豆杉药材的前处理步骤,节约了实验时间,适用于红豆杉枝叶中紫杉醇含量测定;同时也证明中国思茅地区人工种植的红豆杉具备提取紫杉醇的价值,有较高的开发利用价值。

Abstract:ObjectiveTo develop a method for measurement of paclitaxel in the taxus from Yunnan Province. MethodsThe medicinal plants was extracted with ethanol and dichloromethane. ResultsThe theoretical plate number of paclitaxel was 5628, the resolution was 1. 42 and the tailing factor was 1. 01. The calibration curve was linear in the range of 2

The limit of detection was μ g/ml,the recovery rate was %,%,% and RSD was %,%,% (n=5) respectively for paclitaxel. ConclusionOur developed method can shorten the time used for pretreatment, and is a simple and reliable method to determine the content of paclitaxel in taxus. The results also prove that the quality of yew in Yunnan Province is relatively good.
