摘要:阿拉伯半乳糖蛋白(arabinogalactan proteins, AGPs)是一类广泛分布于高等植物中高
度糖基化的蛋白,在植物的生长发育过程中发挥着重要作用。拟南芥 AtAGP11 基因在雄蕊、
花粉和花粉管中呈现出特异性的表达模式。为进一步了解 AtAGP11 基因在植物生长发育过
程中的可能作用,本文运用花粉和胚胎的分离技术、DAPI 染色技术、人工授粉杂交技术和
半薄切片技术,并结合生物统计学方法对 AtAGP11 超表达转基因植株进行表型观察,发现
在 AtAGP11 超表达植株中存在着较为严重的败育现象。进一步的研究结果显示,AtAGP11
超表达抑制了花丝细胞的生长,引起花丝变短,导致花粉无法授于柱头上而产生植株败育。
本研究发现过量表达 AtAGP11 基因影响了花丝细胞的正常生长,这表明该基因可能在细胞
生长过程中起着重要的作用。

Over-expression of AtAGP11 gene leads to abortion in
Arabidopsis thaliana

Abstract: Arabinogalactan proteins (AGPs) are a kind of highly glycosylated protein, widely
distribute in higher plants and play an important role in plant growth and development process.
AtAGP11 is specially expressed in the stamen, pollen and pollen tube. In order to further
investigate the possible role of AtAGP11 in plant growth and development process, we adopted
pollen and embryo separation, pollen DAPI dyeing, artificial pollination, semi-thin section
technologies and biostatistics to observe the phenotypes of AtAGP11 over-expression transgenic
plants and found serious abortion phenomenon in the AtAGP11 over-expression plants. Further
research showed that over-expression of AtAGP11 affected pollination by shortening the length of
filaments so that pollen could not land on stigma, resulting in abortion phenotype. This study
discovered that over-expression of AtAGP11 affected regular growth of filaments, indicating a
key role of AtAGP11 in cell growth.
