[摘要] 目的观察丹参酮ⅡA对阿霉素所致大鼠心肌损伤的保护作用及可能机制。方法将SD大鼠随机分为对照组、模型组(阿霉素心脏损伤)、丹参酮ⅡA组(×10-5、×10-5、×10-5 mol/L)。丹参酮ⅡA组大鼠造模前给予20 mL/kg体积丹参酮ⅡA混悬液灌胃,连续5 d,第5天给药后1 h腹腔注射18 mg/kg阿霉素溶液,此后继续灌胃2 d。检查大鼠的心脏功能,检测大鼠血浆乳酸脱氢酶(LDH)、肌酸激酶(CK)、丙二醛(MDA)、超氧化物歧化酶(SOD)水平和心肌组织中肌钙蛋白Ⅰ(cTnⅠ)水平。结果与模型组相比,丹参酮ⅡA组(×10-5、×10-5 mol/L)明显改善心脏功能,表现为左室收缩压[(±)mm Hg比(±)、(±)mm Hg,F=]、左室压力差[(±)mm Hg比(±)、(±)mm Hg,F=]、左室内压最大上升速率[(±)mm Hg/s比(±)、(±)mm Hg/s,F=]和左室内压最大下降速率[(±)mm Hg/s比(±)、(±)mm Hg/s,F=]均显著性升高(均P < )。与模型组比较,丹参酮ⅡA(×10-5、×10-5 mol/L)显著降低了心肌组织匀浆中cTnⅠ[(±)ng/mgpr比(±)、(±)ng/mgpr,F=]、LDH[(±)U/L比(±)、(±)U/L,F=]、CK[(±)U/mL比(±)、(±)U/mL,F=]和MDA[(
±)μmol/mL比(±)、(±)μmol/mL,F=]的水平,增加了SOD活性[(±)kU/L比(±)、(±)kU/L,F=](均P < )。结论丹参酮ⅡA对阿霉素诱导的大鼠心肌损伤具有保护作用,其机制可能是丹参酮ⅡA抑制氧化应激有关。
