Abstract: AIM: To observe antihypertensive effects of Apocynum um L. leaf extract(ALE) and explore its part mechanism in 2-kidney, 1-clip (2K1C) rats on high-fat and refined-carbohydrate diet. METHOD: Rats with metobolic syndrome were copied by two-Kidney, one-Clip method and high-fat and refined-carbohydrate diet for twelve weeks. Systolic blood pressure(SBP) was determined at 5th week, forty-two rats with SBP exceeding 160mmHg were randomly assigned to five groups: ALE with high-, middle- and low-dose groups(200, 100, 50mg·kg-1·d-1, n=8), captopril group(30 mg·kg-1·d-1, n=8) and model group(%carboxymethylcellulose 5 ml·kg-1·d-1, n=10), another laying sham-operated group(%carboxymethylcellulose 5 ml·kg-1·d-1, n=10). Rats were intragastric administrated between 9:00-10:00 everyday for eight weeks according to above-mentioned setting dosage. Body weight was recorded weekly, and the dose was adjusted adcording to weight changes. At 5th, 9th and 13th week, SBP and urinary volume for 24 hours were determined. At 13th week, body weight, left clipped kidney weight and white adipose tissue humid weight(WHW) were weighed, oxidative and antioxidative indexes for clipped kidney tissue, levels of angiotensin Ⅱ and aldosterone for plasma, content of insulin, leptin, TNF-α, IL-6, TG, TC, Scr and BUN for serum were measured. Pathological changes of liver tissue was observed by hematoxylin-eosin staining method. RESULTS:ALE treatment significantly reduced established hypertension at 9th and 13th week. The level of oxidativ
