摘要:目的从海水中分离活性较高的琼胶酶产生菌,以制备琼胶酶用于琼胶寡糖的生产,琼胶寡糖具有多种生物活性,在食品、化妆、医药等行业有广阔的应用前景。方法以琼胶为唯一碳源,用稀释涂布平板法进行分离,平板透明圈法进行初筛,摇瓶培养法进行复筛,改进的DNS比色法进行酶活力的测定。结果从海水中分离筛选到18株琼胶降解菌株,经复筛得到一株产酶活力较高的海洋细菌HS0326-601,并对该菌株进行了初步鉴定,结果表明其为革兰氏阴性细菌,菌体呈短杆状。结论鉴于该菌株生长速度快,通过对其进一步的诱变和发酵条件优化,有望成为高产琼胶酶产生菌,从而为琼胶酶、琼胶寡糖的深入研究和开发应用奠定基础。

Isolation, Screening and Identification of Agarase–Producing Bacteria from the Seawater

ABSTRACT: OBJECTIVE To degrade agar into agaro-haride by the agarase with a higher activity, the strains were separated from seawater. The agaro-haride have shown wide application values in food industry, cosmetic industry, pharmaceutical industry and other sectors. METHODS The agarase-producing baceria were isolated by the spread plate method, first-screening by clear halos on agar plate containing agar as the sole-carbon source, second-screening of shaking culture. The enzymatic activities were determined by improved DNS colorimetry method. RESULTS 18 agar-degrading strains were isolated from the seawater, and a more powe
