【摘要】利用Minitab软件,采用响应面方法(response surface methods,RSM)对多杀菌素发酵条件进行优化。通过PlackeetBurman设计实验,从培养基中筛选出显著性影响因子:葡萄糖、棉籽粉、玉米浆的浓度以及初始pH,并确定非显著影响因素的水平;采用中心组合设计,对四个显著影响因素进一步的优化,根据建立的多元二次方程确定其最佳水平,得到最佳的培养条件(g/L):,麦芽糖10,,,,,,CaCO3 5;接种量10%,,摇床转速220r/min,培养7d。在此条件下,多杀菌素的产量为175mg/L,比优化前(120mg/L)%。

ABSTRACT Using response surface methods with the aid of Minitab software, the fermentation materials were investigated and optimized. The significant impact factor of the different fermentation conditions were screened and identified by PlackeetBurman experimental design. The level of significant factors were further optimized using posite design. From the resulting of multiquadratic equation, the optimized levels of influencing factors could be established. ef extract g/L; zinc sulfate g/L; ammonium sulfate g/L; CaCO3 5 g/L and the pH adjusted to . The optimized culture condition: shaker speed 220 r/min; inoculum size 10% v/v; and cultivated for 7 days. The production titer of this condition in shakeflask reached 175 mg/L, which was increased by % compared with the previous condition.
