【摘要】目的:探讨2-(2-苯并呋喃基)-2-咪唑啉(2-BFI)对星形胶质细胞糖氧剥夺(oxygen-glucosedeprivation,OGD)损伤的保护作用及脂质过氧化的影响。方法:制成星形胶质细胞糖氧剥夺损伤模型,分为正常对照组、OGD组、OGD+2-BFI干预组(、、3、6和12μg/mL),测定星形胶质细胞存活率,选出抗OGD损伤最佳浓度,以该浓度进一步观察2-BFI对脂质过氧化产物丙二醛(MDA)和还原型谷胱甘肽(GSH)含量的影响。结果:星形胶质细胞在OGD损伤后,不同浓度的2-BFI均能提高细胞存活率,; 2-BFI干预OGD组能显著降低MDA含量,增加GSH含量。结论:2-BFI能保护星形胶质细胞OGD损伤,且有抑制脂质过氧化的作用。

Abstract: Objective: To explore the neuroprotective effects and lipid peroxidation of 2-(2-benzonfuranyl)-2-imidazoline on oxygen-glucose deprivation of cultured rat corticalastrocytes. Methods: The model of oxygen-glucose deprivation of cultured rat cortical astrocyteswas were designed into three groups: normal control group,oxygen-glucosedeprivation(OGD) group and oxygen-glucose deprivation+2-BFI intervention group (the concentration of drug was ,,3,6 and 12μg/mL),to determine the cell survival rate of astrocytes,the best drug concentration was chosen,then the best drug density was used to measure the content of malondialdehyde(MDA) and reduced glutathione (GSH). Results: When astrocytes were deprived of oxygen and glucose,different concentration of 2-BFI couldsignificantly raised the cell survival rate,and the effect of was the best; g/mL 2-BFI intervention group could significantly reduced MDA level,and raised the content of reduced glutathione. Conclusion: 2-BFI can effectively protect astrocyte injury afteroxygen-glucose deprivation,and inhibit lipid peroxidation.
