【摘要】目的探讨5氨基乙酰丙酸(5aminolevulinic acid, 5ALA)介导的光动力疗法(photodynamic therapy, PDT)对体外培养的人卵巢癌3AO细胞的杀伤效应。方法显微镜观察细胞形态;MTT法检测5ALA介导的PDT(5ALAPDT)对3AO细胞增殖的抑制作用;PI染色及Annexin VPI双染结合流式细胞术分析5ALAPDT对3AO细胞的细胞周期及死亡方式的影响。结果 5ALAPDT后3AO细胞出现典型的凋亡形态学改变;细胞抑制率随着5ALA浓度及激光能量密度的升高而升高(),当光敏剂浓度达到一定水平时,抑制率不再相应升高,该作用存在浓度饱和现象;流式细胞术显示PDT后细胞发生了G0/G1期生长停顿(),有明显的晚期凋亡和继发性坏死()。结论 5ALA对卵巢癌3AO细胞有杀伤作用;凋亡是5ALAPDT杀伤卵巢癌细胞的一种机制。

)ABSTRACT: Objective To study the effects of 5
aminolevulinic acid (5ALA) mediated photodynamic therapy (PDT) on human ovarian carcinoma 3AO cells in vitro. Methods Morphological changes were examined by microscopy; the inhibitory effects of 5aminolevulinic acid mediated photodynamic(5ALAPDT) on 3AO cells were examined by MTT assay; cell cycle of 3AO cells was observed by flow cytometry with PI staining; the modality of 3AO cell death induced by 5ALAPDT was determined by flow cytometry with Annexin VPI staining. Results Typical morphological changes of apoptosis occurred after 5ALAPDT. The inhibition rate of cells increased gradually with the increase of 5ALA concentration and laser irradiation dose (). 5ALAPDT caused G0/G1 phase arrest of 3AO cell cycle (). The late apoptosis and secondary necrosis rates of ovarian carcinoma 3AO cells significantly increased after photodynamic therapy, and they were higher than those of the controls, respectively (). Conclusion 5ALA has a killing effect on ovarian carcinoma 3AO cells; apoptosis might be one of the mechanisms of 5ALAPDT on ovarian carcinoma.

卵巢癌为女性生殖系统三大恶性肿瘤之一,死亡率位居首位。70%的卵巢癌患者就诊时已属疾病晚期,5年存活率只有15%~20%,约有50%~80%的患者将在近期或远期出现复发。光动力疗法(photodynamic therapy, PDT)作为一种新的高选择性肿瘤治疗方法[1],有望成为卵巢癌新的辅助治疗手段。5
