【摘要】目的研究CA 125阴性卵巢上皮性恶性肿瘤患者血清相关蛋白检测及其意义。方法取CA 125阴性卵巢上皮性恶性肿瘤和卵巢良性肿瘤、盆腔良性病变和正常妇女血清,采用双向电泳技术分离和鉴定生理状态与病理状态的差异表达蛋白,对有差异性表达的蛋白质运用MALDITOFTOF串联质谱分析。结果重复性好的双向电泳图谱在CA 125阴性卵巢上皮性恶性肿瘤患者血清蛋白质表达谱发生了改变。将其中有差异蛋白点经指纹图谱(PMF)分析,并结合双向电泳相应点的表观等电点、分子质量、匹配片段的多少以及相关文献进行综合分析。得到CA 125阴性时的蛋白质有8种,分别为结合珠蛋白α1、prostasin、calgranulin A和calgranulin B、溶血磷脂酸(LPA)、增殖细胞核抗原(PCNA)、HSP 60和90。结论双向电泳质谱法对卵巢上皮性恶性肿瘤患者血清蛋白获得的结果重复性较好。CA 125阴性卵巢癌患者血清结合珠蛋白α1、prostasin、calgranulin A和calgranulin B、LPA、PCNA、HSP 60和90能为其提供早期诊断指标。
