【摘要】目的研究联合应用二甲双胍能否增强化疗药物5-氟尿嘧啶(5-FU)对人结肠癌细胞株SW620的抑制作用。方法体外培养人结肠癌细胞株SW620,以5-FU联合或不联合二甲双胍干预细胞,MTT法检测细胞增殖情况,流式细胞仪检测细胞周期及凋亡以及 CD133+ 细胞比例。结果二甲双胍能够增强5-FU对细胞增殖的抑制作用,5-FU联合二甲双胍干预的细胞与单独应用5-FU干预的细胞相比,细胞周期S期比例降低,早期及晚期凋亡率增加,CD133+细胞比例降低。结论二甲双胍与5-FU联合应用对人结肠癌细胞具有协同抑制作用。

【Abstract】 Objective To investigate whether bination of metformin can enhance the inhibitory effect on human colon cancer cell line SW620 of the chemotherapy drug 5-fluorouracil (5-FU). Methods SW620 cells were treated with 5-FU alone or bination with metformin. The proliferation of the cells was assessed by MTT assay, and the cell cycle, apoptosis rate and the sensitivity of CD133 + cells changes were analyzed by flow cytometry. Results Metformin can enhance the inhibitory effect of 5-FU on cell proliferation. Compared with the cells treated with 5-FU alone , the proportion of cells in S phase and CD133+ cells was reduced and apoptosis induction was increased. Conclusions Metformin and 5-FU have a synergistic anti-proliferation effect in human colon cells.
