【摘要】目的: 观察阿魏酸钠(SF)对高糖诱导系膜细胞(GMC)表达PAI-1蛋白和细胞信号转导分子p38MAPK的影响。方法: 原代培养大鼠肾小球系膜细胞,分为正常组、甘露醇组、高糖组、高糖加不同浓度SF组,分别于处理后24 h、48 h收集细胞。用免疫细胞化学检测各组细胞p38MAPK的表达,流式细胞术检测PAI-1蛋白。结果: 48 h内,高糖促进PAI-1和p38MAPK蛋白的表达,而SF能明显抑制高糖的这种作用,其作用随SF浓度的增加和作用时间的延长而加强。结论: SF能拮抗髙糖诱导的GMC表达PAI-1增多和抑制p38MAPK信号通路。

[Abstract] Objective: To observe the influence of sodium ferulate (SF) on PAI-1 and p38MAPK protein expression of mesangial cells in incubation medium with high glucose. Methods: The primarily cultured glomerular mesangial cells (GMCs) were divided into normal glucose group, mannitol group, high glucose group and high glucose plus SF groups of which cells exposed to different concentrations of SF. Cells were collected in 24 h and 48 h after being treated with corresponding agents respectively. The expression of PAI-1 was detected with flow-cytometry and that of p38MAPK protein was detected with immunocytochemistry methods. Results: High glucose significantly promoted the expression of PAI-1 and p38MAPK of GMCs when cultured for 24 h and 48 h. SF inhibited these promotion of PAI-1 and p38MAPK protein induced by high glucose, and the inhibitory effect increased along with the increase of SF concentration and the extension of its affecting time.
