【摘要】目的针对荭草对心血管方面的作用,确定荭草药材的药用部位。方法以大鼠和家兔为实验动物,采用结扎冠脉造成急性心肌缺血和离体心脏冠脉流量灌注模型,分别观察荭草各部位(全株、根及粗茎、带叶茎枝、花穗)对上述实验模型心电图、心肌梗塞范围和冠脉流量的影响;同时对不同药用部位化学成分(异荭草素、荭草素和原儿茶酸)及其指纹图谱进行测定,以探讨荭草对心血管作用的物质基础。结果荭草全株、带叶茎枝、花穗均能不同程度改善缺血心肌心电图,使缺血范围降低;可明显增加离体心脏的冠脉流量。其中带叶茎枝和花穗的综合作用较优,而根及粗茎最弱。不同部位异荭草素、荭草素和原儿茶酸的含量均呈现相同趋势:带叶茎枝>花穗>根及粗茎;指纹图谱亦显示不同药用部位的特异性差异。结论荭草的带叶茎枝和花穗可能是心血管疾病治疗的有效药用部位,并且其中异荭草素、荭草素等化学成分的多寡与药效强度具有一定相关性,可为药材的临床应用和质量控制提供依据。

Abstract:ObjectiveTo find out the effect of medicinal parts of Polygonum orientale on cardiovascular models of acute myocardial ischemia induced by ligating coronary artery in rats and coronary artery flow(CF) in isolated heart
of rabbits were applied. The effects of different medicinal parts(including the whole plant,root & main stem,branch with leaves, ear) on the ECG, the infarct sizes and CF of heart were contents of chemical constituents(isorientin, orientin,protocatechuic acid) and HPLC fingerprint were determined in order to find out the substance basis in cardiovascular disease extracts from whole plant,root & main stem,branch with leaves and ear of the plant could improve the change of ECG, decrease the range of myocardial ischemia and increase the CF to varying degrees.
