摘要:目的研究灵芝孢子粉对裸鼠移植性人肝肿瘤血管生成的影响,并初步探讨其抑瘤机制。方法建立裸鼠移植性人肝肿瘤模型,采用随机区组设计法随机分为空白对照组、阳性对照组及灵芝孢子粉组,观察裸鼠肿瘤生长。采用苏木精-伊红染色法、SP免疫组织化学法及病理Imagepro (VEGF)和微血管密度(MVD)。,%,与空白对照组比较有显著差异(P<)。镜下观察,灵芝孢子粉组与空白对照组相比,肿瘤坏死组织较多,细胞异型性较小。灵芝孢子粉组VEGF、MVD表达水平较空白对照组有明显下降。结论高浓度及大剂量的灵芝孢子粉对肝肿瘤有明显抑制作用,并能抑制肿瘤新生血管生成,其机制可能与对VEGF表达的抑制有关。

Abstract:Objective To evaluate the inhibitory effect and ic effect of Lucid Ganoderma spore(LGS)on the growth of the human hepatocarcinoma HepG 2 xenograft in athymic nude The nude mice models of human hepa-tocarcinoma HepG 2 were establishe
d and randomly divided into a NS blank control group,a5-FU positive treatment control group and a LGS treatment -E staining,SP immunohistochemistry and quantitative pathologic image analysis system were used to investigate the microvessel density(MVD)and expression of vascular endothelial growth factor(VEGF)in tumor ti- The in vivo study revealed that LGS produced quite certain therapeutic effect on the growth of the hepatocarcino-ma cell strain(dose:,inhibitory rate:%,P<).Morphologic study showed there were more necrosis and less degree of atypia,decreased MVD and VEGF expression in the LGS group,pared with that in the blank control LGS could inhibit the growth and angiogenesis of the transplanted tumor tissue of human hepatocarcinoma HepG 2 in nude mechanisms may be related to the reduction of VEGF expression.

大量的实验研究表明,肿瘤新生血管的形成与肿瘤的生长和转移扩散密切相关[1] ,肿瘤的血管密度与实体瘤的预后有关。抑制新生血管的形成,则能抑制肿瘤的生长和转移,使肿瘤处于休眠状态[2] 。灵芝是担子菌纲多孔科灵芝属真菌,其生长成熟后所散发出来的孢子体即灵芝孢子粉。灵芝孢子粉富含三萜、多糖及微量元素等有效成分。临床报道孢子粉可改善机体的免疫功能,防治癌症,减轻患者对放疗、化疗的不良反应[3~5] 。近来有报道显示,灵芝提取物对前列腺癌细胞的血管内皮生长因子(VEGF)和肿瘤生长因子
β 1 (TGF-β1 )有抑制作用[6] 。为了进一步证实其客观疗效并研究其作用机制,本实验选用裸小鼠人肝癌He
