【摘要】目的观察枸杞子提取液(LBA)对RCS大鼠视网膜色素变性的治疗作用。方法 RCS大鼠24只,随机分为给药组和对照组,各12只。给药组大鼠出生后10 d开始喂食LBA(1 mg/kg), 对照组大鼠喂食等量生理盐水。两组大鼠分别于生后25、35、45 d处死,取出眼球,制备组织切片,苏木精-伊红染色和TUNEL法检测视网膜感光细胞的数量。结果与对照组比较,给药组大鼠出生25 d时感光细胞数目增多,TUNEL阳性细胞数目减少,差异有统计学意义(t=、,P<)。结论在RCS大鼠视网膜变性的早期应用LBA,可对早期变性的神经元发挥保护作用。

[ABSTRACT] Objective To observe the therapeutic effect of LBA on the hereditary retinal dystrophy RCS rats. MethodsA total of 24 RCS rats were evenly divided into treatment and control group in random. Starting from 10 postnatal days, LBA1 mg/kg was fed on in the treatment group and saline on the control. Photoreceptor necrosis and apoptosis were assessed by HE staining and TUNEL detection. pared with the control, at 25 postnatal days, the number of photoreceptor was more than that of the control (t=,P<), the number of TUNEL positive cells decreased (t=,P<). Conclusion Early use
of LBA is of protective action on early stage of retinal degeneration.

视网膜色素变性是眼科十分常见的一种致盲性疾病,目前全世界约有数百万病人,在我国其发病率呈逐年上升趋势。RCS大鼠是一种自发性蜕变的大鼠,视网膜色素上皮不能有效吞噬感光细胞外节脱落的膜盘,致使膜盘堆积,感光细胞变性[1,2],为研究视网膜色素变性的经典动物模型。 LAVAIL等[3,4]研究显示,RCS大鼠视网膜色素变性过程中感光细胞处于低氧状态,高浓度氧可延缓感光细胞的凋亡,而氧不足则加速感光细胞的凋亡。枸杞子是茄科植物宁夏枸杞(Lycium barbarum L.)的成熟干燥果实,为常用中药,具有滋补肝肾、益精明目之功效[5]。有研究结果显示,枸杞子对青光眼的节细胞(RGCs)有神经保护作用[5]。然而,以往研究结果缺乏广泛的动物病理模
