摘要:在基于热水浴-Triton X-114 相分离特化空间蛋白质的基础上,基于比较蛋白质组学
方法和生物信息学分析,对在 Acidithiobacillus ferrooxidans 在元素硫和亚铁中培养的
细胞胞外、外膜和周质空间蛋白质进行研究,筛选与硫氧化密切相关关键蛋白。获得了 39
个与硫氧化相关的蛋白质,其中含有半胱氨酸残基的蛋白质约占 70%,并有部分蛋白质中半
胱氨酸丰度较高,含有一个到两个 CXXC 结构域。半胱氨酸残基上的巯基在硫氧化过程中具
有重要活性功能,位于细菌细胞包被中这些富含半胱氨酸的蛋白质的鉴出,表明这些蛋白质
在元素硫的活化/氧化过程中具有重要作用。

Abstract: Based on water-bathing-Triton X-114 phase separation of the partment
-specific proteins, and parative proteomics and bioinformatic analysis, the extracullular,
out-membrane and perplasmic space proteins were studied to isolate the key proteins reltating to
the sulfur oxidation of Acidithiobcillus ferrooxidans 23270 that were cultured on substrates
elemental sulfur and ferrous iron, respectively. 39 sulfur oxidation related proteins were isolated
and identified. 70% of which contain cysteine residues, some have higher cysterine content and
one to two CXXC strucutral domans. Thio group of cysteine has important function in sulfur
oxidation, the identification of the cell envelpe proteins rich-in cysteine indicating these proteins
have key role in activation and oxidation of elemental sulfur.
