dimethylaminohydrolase 2 DDAH2)在糖尿病脓毒症机体表达变化探讨血管内皮功能损害发

生脓毒症组)。HE 染色及免疫组化方法观察肺脏组织结构变化及 DDAH2 表达分布变化。
Griess 法测定血清及肺组织一氧化氮(nitrite oxide,NO)含量,Real-time PCR 测定肺组织
DDAH2 mRNA 水平。结果糖尿病组大鼠肺组织 DDAH2 表达减少, RT-PCR 测定 mRNA
表达水平低于正常对照组(P<);免疫组化显示平均光密度值(AOV)无明显变化,脂多
糖(harides,LPS)腹腔注射后正常组大鼠及糖尿病组肺组织 DDAH2 AOV 值
及 mRNA 表达较正常对照组均明显降低(均 P<);糖尿病组 LPS 注射后 AOV 值及 DDAH2
mRNA 水平较基础值更为降低(P<),前者 D 组低于 B 组(P<),后者 D 组低于
B 组(P<)。糖尿病脓毒症组及单纯脓毒症组一氧化氮水平均增高,但前者低于后者,
(P<)。在肺组织中表达,两组均显示出高 NO 水平,且前者组近 2 倍高于后者,(P<
)。结论脓毒症肺脏表现为表达 DDAH2 下调,糖尿病脓毒症肺组织表现为 DDAH2
表达更为明显下调,提示更为严重血管内皮细胞损害,因此 DDAH2 表达下调可能参与脓毒
症糖尿病机体肺脏损伤的发生。

Abstract: [ABSTRACT] AIM: To investigate the role of DDAH2 in endothelial damage of septic
diabetes host. METHODS: Sixty-four wistar rats were randomly divided into 4 groups : namely
normal control group(group A), diabetes group(group B),sepsis group(group C),diabetes sepsis
group(group D ).The loctical expression of DDAH2 was measured by immunocytochemical
mRNA expression of DDAH2 was detected by real-time PCR . quantify the NO
release with Griess method. RESULTS: In diabetes rats , immunocytochemical staining showed
no obvious decreased AOV level(P>), RT-PCR revealed decreased expression of DDAH2
mRNA (P<),compared with control group. LPS injection . induced more lower level
expression of DDAH2 ( P< ) ,AOV
