摘要
摘要:本设计是为了研究膨胀石墨在植物组织培养中的应用。以H2SO4和HNO3的混酸作为酸化剂,KMnO4做氧化剂,采用化学氧化的方法制备可膨胀石墨,在1000℃的温度下膨化,制得膨胀石墨。通过将不同粒度(+32目。+100目,-100~+160目)和不同含硫量的膨胀石墨加入到植物组织培养的培养基中,通过对照实验和正交实验法,将不同粒度和含硫量的膨胀石墨加入到培养基中,并在不同的培养基条件(膨胀石墨粒度、含硫量、膨胀石墨用量、生长素浓度)接种相同的植物组织,根据对照实验方案,在相同的培养基和组织在不同的条件(理想条件和自然条件)下培养,在培养一个月之后的结果,来判断膨胀石墨在组织培养中的作用和效果。
~。发现在苏铁组织培养中,具有很好的抑制细菌生长的作用,未加石墨的染菌几率是加入石墨的5倍。并能促进苏铁愈伤组织的形成和抑制褐化现象的发生,%形成愈伤组织,而未加石墨的全部染菌。在月季的组织培养中,对月季的成活率和生长有明显的促进作用,加入石墨的发芽率是未加石墨发芽率的3倍,并且加入石墨后的月季组织生长较好的占65%以上,是未加石墨的5倍。因此,由以上两种植物组织培养的实验结果证明,膨胀石墨可以应用到植物组织培养中。并且对植物组织培养是有利的,不仅具有良好的抑制细菌生长的作用和抑制褐化现象的发生,而且在一些技术比较成熟的植物组织培养中(如月季)对成活率和生长的促进作用较明显。

ABSTRACT
This design is made to research the use of expansion graphite in the culture of plant can get expansion graphite under 1000℃ by using the mixture of H2SO4 of HNO3 as acide,KMnO4 as oxidizing agent and using chemical oxidization .through putting different size (+32mu,+100mu,-100~+160mu)and sulphur conten expomision graphite into culture medium of plant tissue ;through contrasting experiment and perpendicular experiment, and put different size and sulphur content expansion graphite into culture medium;and under the conditions of different culture medium(the size of expansion graphite, sulphur content, the volume of expansion graphite, growth hrmone concentration)to moculate the same plant to the condition natural condition,and in the process of should observe and write down the relative tada, and we can judge the effect and point of expansion graphite used in the culture of tissue, through the result of one month’s of culture.
Through the experiments, We find that is to sevy we should put ~ expansion graphite into per milliliter culture medium. We found that in the cycad tissue,it can restrain the growth of germ well, and the culture without graphite is time them the culture with it. And the graphite can promote the forming
