大体标本冰冻薄层断面切片机的研制

提要:目的制备大体冰冻薄层断面标本,为三维重建提供丰富、准确的原始数据。方法在冰冻切片技术的基础上设计
和自制大体冰冻软组织切片机,改进了相关的冰冻技术,在冰盒内通过定标板埋设定标线,利用螺旋升降系统调节断层厚度。结果
断层厚度小于 0. 5 mm(本实验为 0. 865 mm) 。并解决了冰冻标本的定标问题。结论该切片机操作简单、经济、实用,获得的图
像清晰、定位准确,且实验周期短,在生物组织三维重建中具有广阔的应用前景。

Preparation of cryomicrotome for thin cross sections of gross specimen

Abstract : Objective To prepare cross sections of frozen gross specimen to provide adequate original data for the three dimensional reestablish
ment. Methods Based on the frozen section method , the frozen soft tissue microtome was designed and prepared. Some relevant frozen techniques
were improved. The calibration line was buried in the ice box with the use of calibration board so that the thickness of the section could be adjusted
by the spiral ascending and descending system. Results The thickness of the section was thinner than 0. 9 mm (0. 865 mm in our experiment) .
The problem of calibration of the frozen specimen was solved. Conclus
