摘要:以乌拉尔甘草不一样愈伤组织为材料,采取石蜡切片和半薄切片技术,对乌拉尔甘草不一样愈伤组织细胞进行了观察,结果表明,半薄切片在制作不一样愈伤组织装片时优于石蜡切片,其清楚度高,分辨率更为准确。石蜡切片中,根源愈伤组织有核细胞较少,而且细胞出现解表现象;半薄切片中观察到了叶源愈伤组织细胞中的着色颗粒、线粒体和圆球体,可为透射电镜制样提供有效的材料。

Comparison of Paraffin Section and Semi-Thin Section of Glycyrrhiza uralensis Callus

Abstract: Cells from different sources of Glycyrrhiza uralensis Fich. callus were observed using paraffin section and semi-thin section technology taking callus from different sources of G. uralensis as trial material. The results showed that semi-thin section which had higher resolution and clarity in making section from different sources of callus. In paraffin section, callus from G. uralensis roots had few cells with nucleus and the cells presented the disintegration phenomenon; Colored particles, mitochondria and spherosomes in the cells in semi-thin section could be observed clearly, which could provide effective material for transmission electron microscope.
