黄颡鱼心脏中分离到1株细菌GM2402,对该菌株进行分离培养、形态学观察、生理生化试验与16S rDNA基因的PCR扩增及序列分析。结果表明,菌株GM2402为革兰氏阴性杆菌,具动力性,V-P试验、水杨酸、过氧化氢酶、脲酶等阳性,不发酵鼠李糖、乳糖等,硫化氢、枸椽酸盐等阴性。16S rDNA序列长1417bp,采用什么方法获得的此结果要说明。采用PCR方法进行体外扩增
GenBank中的登录号是JX101598,且与数据库中小肠结肠炎耶尔森氏菌(Yersinia enterocolitica)%,系统发育树显示与小肠结肠炎耶尔森氏菌聚为一个分支。综合菌株GM2402的生化特性及分子生物学分析结果,判定菌株GM2402为小肠结肠炎耶尔森氏菌,为水生动物致病菌的分子鉴定积累了参考资料。

One bacteria strain named GM2402 was isolated from the heart of natural diseased yellow cartfish (Pelteobagrus fulvidraco), the strain was identified according to its morphological characteristics, physiology and biochemistry experiments, as well as, PCR amplification of 16S rDNA genes and sequence analysis. The results showed that strain GM2402 was Gram-negative bacterium, the ability to movement and MR-VP, salicylic acid, catalase and urease were positive, but L-rhamnose, Galactose, H2S and citrate salt were negative. 16S rDNA sequence of strain GM2402 was 1417bp and accession number was JX101598. Strain GM2402 shared % identity with Yersinia enterocolitica strains in GenBank database. ic tree demonstrated that strain GM2402 was gathered into one cluster with Y. enterocolitica. Both biochemical character and molecular analysis were proved that strain GM2402 was Y. enterocolitica. It will be benefited for the identification of Y. enterocolitica in aquatic animals.
