1927年，Joslin在美国临床研究会19届年会报道中说：
“随着饮食治疗和胰岛素的应用，现今糖尿病治疗的到了明显的改观，在这种情况下，要想评价其他任何一种新的治疗手段的价值都极其困难。”
“更为重要的是，对双胍类化合物降糖效果的研究为今后可能发现更多的其他口服降糖药奠定了基础，在今后，口服降糖药会在某种程度上取代胰岛素。”
“The modern diabetic with diet and insulin makes it exceedingly difficult to estimate the value of any new method of treatment.”

磺酰脲类的“意外低血糖事件”
二战时期，传染病促使人们大量使用磺***类药物。Marcel Janbon在临床使用2254RP时，发现患者出现强烈的惊厥发作，称之为“意外的低血糖事件”。但由于该药的毒副作用，使磺脲类药物到战后才进入临床
双胍类药物的“降糖副作用”
同时，Curd同样开发出一种具有双胍结构的抗疟疾药物，而其衍生物——二甲双胍也在南非有效地用于治疗流感。当时便已发现，双胍类可以影响血糖但不会引起严重的毒副作用
