摘要:对四川、广西、海南、浙江4地的八角属植物中莽草酸的含量进行了测定,得到广西产八角茴香(Illicium verum Hook. f.)中莽草酸含量最高,%。对莽草酸的两种提取工艺进行了比较,得到水提取工艺效果较好,有利于莽草酸的后续纯化,最佳工艺为按水料质量比10∶ h,提取3次。莽草酸的纯化工艺有萃取法、阳离子树脂交换法、阴离子树脂交换法、硅胶吸附法等,其各有利弊,综合分析以树脂交换法为佳。

Study on the Extraction and Purification Craft of Shikimic Acid from
Illicium verum Hook. f.

Abstract: The shikimic acid content in Illicium plants from Sichuan, Guangxi, Hainan, Zhejiang were measured. The results showed that star anise(Illicium verum Hook. f.) from Guangxi had the highest shikimic acid content, which was %. Two extraction process for shikimic acid were compared, and it was concluded that water extraction method had better extraction efficiency and was benefit for later purification of shikimic acid. The optimum craft was, mass ratio of material to water at 10∶1 and reflux extracting for h. Methods for shikimic acid purification included extraction method, cation exchange resin method, anion exchange resin method, silica gel adsorption method and so on. Each method had advantages and disadvantages. According to comprehensive analysis, resin exchange method was better.

莽草酸(Shikimic acid)化学名称为3,4,5-三羟基-羧酸,白色粉末,分子式为C7H10O5,溶于水、甲醇、乙醇、正丁醇,几乎不溶于石油醚、***、***仿,水中溶解度为180 g/L,提取自八角茴香。中药八角茴香别名大茴香、大料、八角大香,为木兰科植物八角茴香(Illicium verum Hook. f.)的果实,分布于福建、广东、广西、贵州、云南等省区,味辛、温,功能温中理气、健胃止呕,主治呕吐、腹胀、疝气痛等。它不仅是东方的传统调味品,也是法国绿茴香酒和茴香甜酒的主要原料,一般还可增加糖果、茴香酒、烟草的香味。八角茴香是我国南方重要的“药食同源”经济树种,为我国特产香料和中药[1-3]。莽草酸具有很好的抗炎、镇痛和抑制血小板聚集的作用,能抗血栓,抑制动静脉血栓形成[4],是合成抗H5N1型禽流感药物“达菲”的原料[5],自2005年土耳其和罗马尼亚发现H5N1型高致病性禽流感以来,全球掀起一股抢购“达菲”的热潮,作为“达菲”的合成原料莽草酸更是在国际市场价格猛涨,具有广阔的开发价值和市场应用前景。
