文档介绍:杨丰H兴:绿头野鸭鸭疫单默氏杆菌的分离签定及白场疫苗的初步心用绿头野鸭鸭疫里默氏杆菌的分离鉴定及自场疫苗的初步应用研究生:杨祖兴导师:彭大新教授王俊彪高级兽医师摘要鸭疫里默氏杆菌病是由鸭疫里默氏杆菌(Riemerla anatipestife,,RA)引起的, 主要侵害雏鸭、雏鹅和雏火鸡等多种禽类的急性或慢性的接触性传染病。该病能引起较高死亡率和淘汰率,并常与其他鸭的传染病混合感染,给养鸭业造成严重的经济损失。本文从拉萨市林周县某养鸭场的发病绿头野鸭中鉴定出鸭疫里默氏菌,并利用该菌研制了自场灭活疫苗,使用后取得了较好预防效果,为鸭疫里默氏杆菌病的防制起到了参考作用。从发病死亡的绿头野鸭上分离到一株细菌,该菌为革兰氏阴性小杆菌,无芽孢,不运动。菌体呈短杆状,多数菌体单个存在,少数呈双或成短链状排列。在普通琼脂上生长不良,在胰酶大豆琼脂、巧克力琼脂和血液琼脂培养基上生长良好。在厌氧条件下,于血液琼脂平板上37"C培养24小时,形成直径2—、圆形、凸起、边缘光滑、闪光的菌落。该菌不发酵葡萄糖、麦芽糖、甘露糖、乳糖、蔗糖、阿拉伯糖;不液化明胶,不产生硫化氢,不还原硝酸盐,枸橼酸盐利用阴性;能产生吲哚,过氧化氢酶阳性。根据细菌形态、培养特性及生化特性, 鉴定其为鸭疫里默氏杆菌。采用鲜血琼脂平板纸片扩散法进行了该分离株的药物敏感性试验,结果表明该细菌对新霉素、环丙沙星、强力霉素、氟苯尼考、罗美沙星高度敏感;对盐酸林可霉素、先锋霉素Ⅳ、阿奇霉素中度敏感,对复方新诺明、卡那霉素、氟哌酸、四环素低度敏感,而对阿莫西林、磺胺异嗯唑、先锋霉素、磺胺嘧啶、:学位论义敏感。以氟苯尼考注射治疗,效果明显。动物回归试验表明,鸭疫里默氏杆菌对绿头野鸭有较强的致病性,其发病特征与自然发病的症状相似。通过毒力测定,×106CFU。以该菌研制灭活疫苗,免疫后攻毒保护率达到95—100%;在发病的绿头野鸭场使用, %,免疫保护效果良好。关键词:绿头野鸭,鸭疫里默氏杆菌,分离鉴定,疫苗杨祖兴:绿头野鸭鸭疫单默氏杆菌的分离签定及臼场疫苗的初步心用 3 Isolation and Identification ofRiemerella Anatipestifer ingreen’_。head mallards and preliminary application ofautogenous ine :Zuxing Yang Advisor:Daxin Peng Junbiao Wang Abstract Riemerella anatipestifer infectionis allacute orchronic contiguous diseasecaused by hasmainly beenreported inyoung ducks,geese,turkeys,and other ,.infection causes highermortalities,increased condemnations atslaughter,and coinfection withotherduck’S infectiousdisease,which resultsinsubstantial economic losses mercial duckindustry in our thisstudy,we isolated andidentified ,strain from agreen—head mallard farminLinzhou county ofLhasa inTibet,and developed anautogenous killed ineagainstR。anatipestifer study willbehelpful fortheprevention and control ,-infection.
