Codon Usage Bias Analysis of Chloroplast Genome in Camellia
nitidissima

Abstract In order to reveal the codon usage bias and to screen the optimal codon suitable for Camellia
nitidissima chloroplast expression, the codon usage patterns of 52 coding genes (coding sequence, CDS) in
Camellia nitidissima chloroplast genome are studied by using software such as CodonW, Microsoft and R
language. The results show that the use of synonymous codons in the chloroplast genome of Camellia nitidissima
preferred to end with A/U, especially with U. The effective number of codon (ENC) is greater than 35, which is a
weak preference; the base composition of codon position 3 is significantly different from positions 1 and 2, and
the base composition of this position have an important influence on codon usage bias. In addition to natural
selection, codon usage bias is also influenced by mutational pressure and base composition; 17 codons are
identified as optimal codons in the chloroplast genome of Camellia nitidissima. This study lays the foundation for
the genetic modification and phylogenetic study of the chloroplast genome of Camellia nitidissima.
