文档介绍:1 定喘颗粒毒理学研究作者:肖小华,朱令元,徐丽瑛,丁琦,郭晟,方铝【摘要】目的观察小鼠和大鼠分别灌胃给予定喘颗粒的急性毒性作用和长期毒性作用。方法采用 Bliss 法测定小鼠灌胃定喘颗粒的 LD50 以及大鼠连续灌胃定喘颗粒 60 d ,观察给药 60d 的生长、发育,血液学,血液生化学,组织病理变化及停药 21d 时上述指标的变化。结果昆明种小鼠灌胃定喘颗粒的 LD50 为 g/kg 。定喘颗粒大鼠给药 60 d 的毒性试验, 剂量分别为 , , g/kg ( 指含生药量,下同) (相当于临床 50 kg 人用量的 , 和 13. 2 倍) ,给药期间各组大鼠毛色、活动、二便均无异常变化, 但给药组大鼠摄食、饮水、体重以及部分血液学、血液生化学指标和脏器系数有异常变化, 停药后, 上述异常变化指标均已恢复正常。给药期和恢复期重要脏器病理组织检查均未发现明显异常变化。结论定喘颗粒小鼠灌胃的 LD50 为 g/kg ;长期毒性研究表明,长期大剂量灌胃给予定喘颗粒对大鼠有一定毒性,但停药后均已恢复。 2 【关键词】定喘颗粒急性毒性实验长期毒性实验 Abstract:Objective To observe the acute toxicity of Dingchuan granule (DCG) in mice and the long-period toxicity in rats, of DCG were tested in mice by Bliss were administered with repeated gastric infusion of DCG for 60 days and body wEight,hematological and hematobiochemical parameters,coefficient and histomorphological figure ans were examined in 60 days and 21 days after the cessation of DCG. ResultsThe LD50 of KM mice was .In the long-period toxicity test, DCG was administered repeatedly for 60 days, the dose were , , g/kg respectively(It refered to the dose of material,which amounted to , , times of 50 kg human clinic use). There is no other abnormal changes about color patem,action,urine and stool,but cibation, hydroposia, body wEIght and index part of hematological and hematobiochemical parameters,coefficient and histomorphological figure ans were examined during administration rats were examined in 21 days after the cessation of DCG, the abnormal changes of index pletely recovered. After administered repeatedly for 60 days 3 and in 21 days after stopping the drug, there had no obvious abnormality for the an pathology examination. ConclusionThe LD50 of mice is g/kg.
