菌种质量控制
在生产之前，应对所用菌种进行检查，确认其纯度和应用性能没有发生退化。出现污染或退化的菌种不能作为生产用菌种，。

菌种不纯时，应进行纯化。可采用平板划线分离法或稀释分离法，得到纯菌种。必要时可采用显微操作单细胞分离器进行菌种分离纯化。
对纯化的菌种应进行生产性能的检查。
菌种的复壮
菌种发生下列现象之一，应进行菌种复壮：
—— 菌体形态及菌落形态发生变化；
—— 代谢活性降低，发酵周期改变；
—— 重要功能性物质的产生能力下降；
—— 其他重要特性的退化或丧失；
菌种复壮方法：回接到原宿主或原分离环境传代培养，重新分离该菌种。
发酵增殖
种子扩培
原菌种应连续转接活化至生长旺盛后方可应用。
种子扩培过程包括试管斜面菌种、摇瓶（或固体种子培养瓶）、种子罐发酵（或种子固体发酵）培养三个阶段，操作过程要保证菌种不被污染、生长旺盛。
培养基
培养基重要原料应满足一定的质量要求，包括成分、含量、有效期以及产地等。对新使用的发酵原料需经摇瓶试验或小型发酵罐试验后方可用于发酵生产。
